ABSTRACT

Mycobacterium avium subsp. paratuberculosis, the causative agent of Johne’s disease is a degenerative chronic granulomatous disease of bovines. In the present study, quantitative real time polymerase chain reaction (qRT-PCR) using TaqMan chemistry targeting the IS900 sequence of Mycobacterium avium subsp. paratuberculosis (MAP) was employed for the molecular diagnosis of the disease in bovine faecal samples. Out of 200 bovine faecal samples processed, 7 samples were tested as positive by IS900 qRT-PCR. The sensitivity limit of detection of MAP DNA in faecal samples by qRT-PCR TaqMan assay was found to be 0.05pg. No amplification was observed in other Mycobacterial spp. viz. M. phlei, M. smegmatis, M. intracellulare and M. kansasii.

ABSTRACT

Pasteurella multocida [P. multocida] is an important pathogen of various domestic animals. The outer membrane proteins [OMPs] play a major role in pathogenesis and immunogenicity of P. multocida. The aim of the study was to develop indirect enzyme linked immuno sorbant assay [ELISA] based on OMPs to ascertain the antibody titers in animals post-infection or to gauge the potency of vaccine. The OMPs were extracted and purified from P. multocida P: 52 [vaccine strain] and P. multocida B: 2 isolated from natural outbreak of Haemorrhagic septicaemia [HS] and analyzed on SDS PAGE and through western blot. The OMPs profile of the vaccine strain and the isolate from the natural outbreak of HS were found to be similar. Optimization of various components viz. coating antigens, anti-species conjugate, etc. were carried out against both anti-P. multocida hyper immune and pre immune serum. Validation of OMP based indirect ELISA assay to measure immune response against P. multocida in bovine revealed 91% diagnostic sensitivity [DSN] and about 100% diagnostic specificity [DSP] at 25% cut off. OMP based indirect ELISA was found to be more specific, but less sensitive as compared to WCL based assay

ABSTRACT

Present paper deals with the findings of drug efficacy of three fruit drugs viz. Emblica officinalis. Terminalia belerica and Terminalia chebula extracted in aqueous, alcohol and ether - against four bacterial species commonly found associated with urinary tract infection [UTI] i.e. Escherichia coli, Klebsiella, Proteus and Pseudomonas species isolated from forty human urine samples. Maximum antimicrobial potency of each fruit drug was recorded for 100% concentrated extract in all the extract types [aqueous, alcoholic and ether] but a declined trend of drug potency was recorded with the dilution factor of the extract in all cases. In general, maximum drug efficacy was recorded in aqueous extract followed by alcoholic and ether extract. Similar trend was also found in the compound formulated drug [Triphala] prepared from above three different drugs. A positive correlation of drug efficacy with drug concentration was clearly found

Continued interest in the study of nickel [II] complexes has been due to a variety of stereo chemical configurations attained by it and the attending complications of interconversions of different structures and also possible equilibria between different geometries. There has been a good deal interest of nickel [II] complexes of thiosemicarbazones surprisingly there are few reports on nickel [II] complexes of their oxygen analogs, i.e. semicarbazide and semicarbazones. ABSTRACT

Gastrointestinal cutaneous fistula can be a threat to life unless carefully handled. Of the cases of G.I.C.F. treated at the Surgical Department of the Central Hospital, Tripoli, Libya, three occurred post-operatively, one after trauma and one after inflammatory duodenal disease [perforated duodenal ulcer]. Two were high output, three low output. In two cases abscess drainage was performed and all the cases were managed by gastric suction, blood transfusion, fluid and electrolyte replacement, antibiotics, hyperalimentation, suction of fistula discharge, skin ointment. A very important role was played by total parenterals nutrition [T.PN.] Two cases healed after two weeks. The same treatment was given to the other three cases for a period of four weeks, during which time preparation for surgical intervention was made. Resection and anastomosis of terminal ilium was performed med in Case I transfer of BI to BII was performed but there was persistent leakage. The patient was re-explored and later developed septic shock and died. Four of the cases were treated successfully but one died
